MushBag Rye Substrate Instructions

Before you begin: clean your workspace

  • Use soap to wash your hands and disinfectant to clean your work area. Make sure your work area is clean and sterile before arranging all materials and beginning the process.
  • Make sure there are no drafts if growing mushrooms.
  • Wash your hands and arms every time before handling the mushroom substrate kit with hand sanitizer or antibacterial soap. As additional protection, use disinfected gloves and a face mask.

Let’s start by inoculating the liquid culture as a first step. Liquid culture is a sterile liquid rich in nutrients. Typically, it is a mixture of water and different sugars. When injected with mushroom spores, the combination has the ability to produce mycelium growth. If you wish to inject spores from a spore syringe directly into the substrate bag, you can skip this step and proceed to the “Inoculation of the Substrate Kit” section.

Prepare the liquid culture

For this step you need a bottle liquid culture or make liquid culture. And a syringe of mushroom spores or a veil of spores.

Wash your hands or use gloves!

  1. Shake the spore bottle or syringe well to make sure all the spores float.
  2. Disinfect the liquid culture injection site and spore vial or syringe with the alcohol swab.
  3. Inject 2.5 ml of spore solution into 10 ml Liquid Culture or 5 ml of spore solution into 25 ml

Colonize the liquid culture

  • Shake the inoculated liquid culture vigorously once a day every day for 10 days

  • Store the inoculated liquid culture somewhere between 23 and 27 degrees Celsius.

The liquid culture is ready when you see a cloud floating in the suspension. It’s mycelium. Your liquid culture is ready, it is called the master culture, if used and stored correctly you can have a lot of mycelium from just a little bit of spores. Important! If the liquid is cloudy, it means there is contamination and the culture is unusable. If you are not using the liquid culture, store it in the refrigerator.

Inoculation of MushBag rye substrate

Arrange all the materials you need on a disinfected workspace. You will need:

  • Mushbag Rye Substrate Bag
  • Mushroom spore syringe or liquid culture with germinated mushroom spores
  • A lighter or alcohol burner
  • Alcoholic preparations
  • Disinfectant spray

Prepare your mushroom spore syringe with 10 ml of spore solution or use the prepared liquid culture (10 -25 ml). This is enough spores to inoculate the Mushbag Rye Mushroom substrate.

  • Locate the injection port on the substrate bag. Disinfect it with an alcohol swab. Don’t touch it again
  • Shake the spore syringe well for 5 to 10 seconds. This will get the spores from the sides and distribute them more evenly throughout the spore solution.
  • Take the syringe and heat the needle red hot with a lighter to make it sterile. Let the needle cool for a few seconds.
  • Inject the syringe needle into the bag. Add all of the mushroom spore solution to the substrate kit. If flow becomes blocked, pull the needle back slightly.
  • Squeeze and shake the bag carefully to spread the spore suspension throughout the substrate.
  • Store the kit in a dark place at a temperature between 23 and 27 degrees

Colonization of Mushbag Rye

  • To let the mycelium grow and colonize the substrate, you should keep the MushBag Rye in a dark space with a temperature between 26 and 28 degrees Celsius.
  • Please note that lower temperatures will cause colonization to stagnate or extend the colonization time of the substrate. Temperatures below 15 degrees will stop mycelium growth.
  • After 3 to 7 days, you will see mycelium begin to grow from where the spores were deposited. The mycelium resembles cotton threads and becomes stringier as it grows and colonizes the mushroom substrate.
  • During this stage, the Mushbag may swell a little.
  • Shaking the bag can accelerate the growth of the mycelium. Do this only after you have noticed that mycelium has grown in a large portion of the bags. Break it up a bit to spread it out and let the mycelium grow further.
  • It takes 2 to 4 weeks for the mycelium to colonize the substrate in the box. No empty spots of substrate should be left.
  • Wait an additional 5 to 7 days to ensure that the interior of the substrate is also colonized by the spores. Now that the mushroom substrate is completely covered with mycelium, the kit is ready to be prepared for fruiting or used as spawn to inoculate more substrate.

Growing from MushBag Rye

The simplest approach to harvesting mushrooms from mushbag rye is to simply open the bag from the top. Create a layer of vermiculite on the surface of the substrate. Add moisture to the bag, then seal it with clothespins or paper clips. When circumstances are favorable, mushrooms can be harvested within a few weeks.
